lt also reacts with the light chains of other pig immunoglobulins. No antibody was detected against non-immunoglobulin
serum proteins.The antibody may cross-react with immunoglobulins from other species.
Store at -20°C. Avoid repeated freeze / thaw cycles.The expiration date may be extended if test results are acceptable for

答：可能的原因有：
1、膜沒有完全均勻濕透,建議 使用100% methanol浸透膜；
2、洗膜不充分，可以增加洗液體積和洗滌次數；
3、電極不平衡或者加樣位置偏斜，可以調整電極和加樣；
4、封閉物用量不足，可以提高封閉物濃度，孵育時保證封閉液完全浸沒轉印膜；
5、封閉物使用不當，比如檢測生物素標記的蛋白時不可用脫脂奶粉封閉；
6、封閉時間不夠，一般情況下室溫37度封閉1小時以上或者4度封閉過夜；
7、抗體非特異性結合，可以降低抗體濃度，減少孵育時間；
8、一抗稀釋度不適宜，可以對抗體進行滴度測試，選擇適宜的抗體稀釋度；
9、一抗孵育的溫度偏高，建議4℃結合過夜；
10、抗體濃度過高或洗滌不夠，建議降低抗體濃度，增加洗滌次數和時間；
11、化學顯色底物過多，建議按說明書加入適量的顯色底物。
答：可能的原因有：
1、膠濃度不對，不同濃度的膠跑出的蛋白條帶的位置可能有所偏差，可以調整濃度；
2、抗體孵育不充分,建議增加抗體濃度，延長孵育時間；
3、酶失活，建議直接將酶和底物進行混合，如果不顯色則說明酶失活了。選擇在有效期內、有活性的酶聯物；
4、目的蛋白存在翻譯后修飾或剪切體，建議查詢相關文獻確定；
5、標本中不含靶蛋白或靶蛋白含量太低，建議設置陽性對照比對結果，增加標本上樣量。
